AbstractThe I148M variant of PNPLA3 is strongly linked to hepatic steatosis. Evidence suggests a gain-of-function role for this I148M mutant, leaving the physiological relevance of wild-type PNPLA3 undefined. Here we show that PNPLA3 selectively degrades triglyceride (TGs) enriched in polyunsaturated fatty acids (PUFAs) both in cultured cells and in mice.
