Our study addresses the limitations of fecal calprotectin detection in diagnosing and monitoring inflammatory bowel disease (IBD). We developed a portable, tube-based CRISPR/Cas assay for point-of-care testing of calprotectin, with a detection range of 1–10,000 ng/ml. Validated in a preclinical rodent model of ulcerative colitis (UC), this assay showed 2–3 times higher calprotectin levels in UC samples compared to healthy controls.
