For technical reasons, researchers have struggled to map complex biological systems and track disease progression at high resolution across the entire mammalian body. Although organ optical transparency has been achieved in mice and human organs, labelling specific structures and cells within these organs has had limited success. Notably, conventional immunoglobulin G antibodies struggle to penetrate deep into tissues and often form aggregates, limiting the success of whole-body imaging.

New Results doi: https://doi.org/10.1101/2023.02.17.528921 ABSTRACTMost diseases involve multiple interconnected physiological systems, but histological evaluation of their pathology is currently limited to small tissue samples. Here, we present wildDISCO, a technology that uses cholesterol extraction to enable deep tissue penetration of standard 150 kDa IgG antibodies in chemically fixed whole mice.
