AbstractKiwifruit is an economically and nutritionally important horticultural fruit crop worldwide. The genomic data of several kiwifruit species have been released, providing an unprecedented opportunity for pan-genome analysis to comprehensively investigate the inter- and intra-species genetic diversity and facilitate utilization for kiwifruit breeding. Here, we generated a kiwifruit super pan-genome using 15 high-quality assemblies of eight Actinidia species.

Seven Cas13 orthologs were codon-optimized and synthesized by Kingsley Company. The pRGEB32-GhU6.7-NPTII vector was linearized using BstBI and XbaI. Subsequently, the full-length Cas13 was ligated into the linearized pRGEB32-GhU6.7-NPTII using the ClonExpress® II One Step Cloning Kit to obtain pRGEB32-GhU6.7-Cas13-NPTII. The tRNA-gRNA transcription unit has been used to effectively enhance crRNA transcription in the CRISPR/Cas system for plant genome editing [47, 75, 76].

The winter wheat cultivar AK58 was used in this study. After being sterilized with 75% ethanol for 45 s, the seeds were soaked in water containing 1.6% H2O2 for 24 h at room temperature and pre-germinated on wet filter paper. The germinated seeds were transferred to soil and grown in a growth chamber with 16 h light and 8 h dark cycle at 22 °C for normal growth and at 4 °C for vernalization treatment. For V0 treatment, plants were grown at 22 °C for 14 days.
