In order to visualize autophagic vesicle transport, rat LC3 cDNA was amplified, fused with the cDNA for the basic red fluorescent protein mScarlet, and finally packed into adeno-associated viral vectors of the serotype 6 (AAV-6). Recombinant AAV particles were produced in transiently transfected HEK293 cells and were purified from the cell lysate by iodixanol step gradient ultracentrifugation followed by heparin affinity chromatography on an Äkta FPLC.
