AbstractStem cell-based embryo models, which recapitulate symmetry breaking, pattern formation and tissue morphogenesis during early development, provide promising experimental tools to study the development of mammalian species, including humans. Despite considerable progress in embryo modelling using cultured stem cells, generating embryo models with high fidelity, efficiency, controllability, and in vivo-like cellular organization and tissue architecture remains challenging.

The first few weeks of a human embryo’s development are shrouded in mystery. Researchers cannot observe embryos that are nestled away in their mother’s womb until they are large enough to be detected by ultrasound, about three weeks after conception. Few human embryos are donated to research, and regulations in many countries prevent those few from being studied beyond a stage equivalent to 14 days of natural development.

AbstractRegion-specific gut spheroids are precursors for gastrointestinal and pulmonary organoids that hold great promise for fundamental studies and translations. However, efficient production of gut spheroids remains challenging due to a lack of control and mechanistic understanding of gut spheroid morphogenesis.
