MATERIALS AND METHODSpX459 (Addgene, 62988) and guide RNA (gRNA)–AAVS1–T2 (Addgene, 41818), pOG44 (Invitrogen, V600520), pHDM-G (PlasmID, EvNO00061606), pHDM-HGPM2 (PlasmID, EvNO00061607), pHDM-tat1B (PlasmID, EvNO00061608), and pRC-CMV-rev1B (PlasmID, EvNO00061616) were obtained from the indicated suppliers. pDEST-hisMBP-AsCpf1-EC (Addgene, 79009) was modified to remove the maltose binding protein (MBP) sequence using standard molecular biology techniques.
