Key PointsQuestion  What is the accuracy of tumor tissue–modified human papillomavirus DNA (TTMV-HPV DNA) liquid biopsy in the diagnosis and surveillance of HPV-associated oropharyngeal squamous cell carcinoma (OPSCC)? Findings  In this cohort study of 399 patients in a contemporary clinical setting, TTMV-HPV DNA testing at the time of OPSCC diagnosis demonstrated excellent sensitivity and specificity of 91.5% and 100%, respectively.
