AbstractVaccine-induced T cells and neutralizing antibodies are essential for protection against SARS-CoV-2. Previously, we demonstrated that an antigen delivery system, pullulan nanogel (PNG), delivers vaccine antigen to lymph node medullary macrophages and thereby enhances the induction of specific CD8+ T cells. In this study, we revealed that medullary macrophage-selective delivery by PNG depends on its binding to a C-type lectin SIGN-R1.

AbstractCurrent KRASG12C (OFF) inhibitors that target inactive GDP-bound KRASG12C cause responses in less than half of patients and these responses are not durable. A class of RASG12C (ON) inhibitors that targets active GTP-bound KRASG12C blocks ERK signaling more potently than the inactive-state inhibitors. Sensitivity to either class of agents is strongly correlated with inhibition of mTORC1 activity.

Extended Data Fig. 1 Scrib mis-localization following KRAS G12C inhibition. a. The NCI-H358 KRAS G12C mutant lung cancer cell line was treated with 500 nM adagrasib for 48 hours. Representative immunofluorescence images of E-cadherin are shown. Scale bar, 10 μm. b. NCI-H358 cells were treated with 1 μM sotorasib at the indicated time points. Representative immunofluorescence images of Scrib are merged with 4’,6-diamidino-2-phenylindole (DAPI). Scale bar, 5 μm. c.
