The mechanisms of many disease-associated variants are uncertain because of limited power to detect their modest effects on gene expression. This study finds that natural selection leads to preferential detection of disease-associated versus expression-associated variants. Mostafavi et al.4 identified 22,119 lead variants from GWAS across 44 UK Biobank phenotypes and 118,996 lead variants from eQTL analysis of protein-coding genes across 38 tissues from the GTEx and eQTLGen studies5,6.
