Participant data was acquired from four separate cohort studies of aging and dementia. The Vanderbilt Memory and Aging Project (VMAP) was utilized as a discovery cohort while the Religious Orders Study (ROS), Rush Memory and Aging Project (MAP), and the Alzheimer’s Disease Neuroimaging Initiative (ADNI) were utilized for replication analysis. VMAP is a longitudinal study focused on the intersection between vascular health and brain aging [24].

Deidentified data were acquired from NDA (collection ID #2021). Several key features of this dataset guided our selection including the availability of Repetitive Behavior Scale-Revised scores on participants from both groups, adequate sample sizes, and a nearly equal number of males and females.

Statistical analysis Three to five independent cell culture preparations were used for all in vitro experiments, and 5–6 animals per group were used for in vivo experiments. Sample size was not determined by statistical method as the number of independent experiments we used was sufficient to detect significant changes as reported previously (Wang et al., 2022). No exclusion criteria were pre-determined. Data of all animals in the experiments were included for analysis.
