AbstractA hallmark of Alzheimer's disease (AD) is the extracellular aggregation of toxic amyloid-beta (Aβ) peptides in form of plaques. Here, we identify netoglitazone, an antidiabetic compound previously tested in humans, as an Aβ aggregation antagonist. Netoglitazone improved cognition and reduced microglia activity in a mouse model of AD.

Immune cells, patrolling through different environments of the body, are a prime example of cell plasticity in response to the environment. Circulating immune cells can alter their step when infiltrating cancer, developing excessive immune tolerance to the tumour and thus allowing cancer cells to grow unchecked. Single-cell genomics has enabled genome-wide molecular characterization of individual cells within the tumour microenvironment (TME) in an unbiased manner.
