H1 male and WIBR3 female human embryonic stem cells were provided by the Krause and Jaenisch lab, respectively, and both tested negative in the Mycoplasmacheck from eurofins Genomics upon receipt and throughout the study. Primed H1 were transduced with GFP or KLF4-containing lentiviral vectors and split after 48 h then selected using blasticydin for the 3 following days.
