In this study, we first demonstrated that CD44 signaling contributes to β-AR-induced activation of RyR2 and PLN through the interaction with Epac1 and activation of CaMKII. Inhibition of CD44 is effective in preventing β-AR-induced Epac1-dependent activation of RyR2 and PLN, which leads to Ca2+ leakage from SR and the subsequent VAs. CD44 is upregulated in HF. Eliminating CD44 is effective in reducing VA inducibility in HF mice.
