The peptide antibiotic teixobactin achieves its remarkable activity through the unique pattern of d-l-l-d-d-l-l stereochemistry in amino acid residues 1–7. (1−3) Teixobactin comprises a macrolactone ring (residues 8–11) and a linear tail (residues 1–7); the ring binds the pyrophosphate anion of lipid II and related bacterial cell wall precursor molecules, and the tail forms an amphiphilic β-strand that induces β-sheet assembly (Figure 1).
