RESEARCH HIGHLIGHT 02 May 2025 Hundreds of mutations in POLG — the gene encoding the catalytic subunit of DNA polymerase γ (POLγ), which replicates mammalian mitochondrial DNA (mtDNA) — are the cause of severe, progressive conditions, with no available treatments. Now, a study in Nature reports the discovery of a small molecule activator of POLγ that boosts mtDNA synthesis in patient-derived fibroblasts harbouring various POLG mutations.
