AbstractMolecular glues are proximity-inducing small molecules that have emerged as an attractive therapeutic approach. However, developing molecular glues remains challenging, requiring innovative mechanistic strategies to stabilize neoprotein interfaces and expedite discovery. Here we unveil a trans-labeling covalent molecular glue mechanism, termed ‘template-assisted covalent modification’.

Graphical Abstract One obstacle to a better understanding of the “dark” lipid kinase PIP4K2C is the lack of chemical tool compounds for this protein. Here, we describe a picomolar PIP4K2C binder TMX-4102, and an effective bivalent PIP4K2C degrader, TMX-4153, both of which display exclusive binding selectivity for PIP4K2C. Thus, TMX-4102 and TMX-4153 represent two useful leads to further interrogate the therapeutic potential of PIP4K2C.
