1 INTRODUCTION Calpains are cytosolic cysteine proteases that function at neutral pH (Ono et al. 2016; Vu et al. 2022; Zatz and Starling 2005). Calpain-1 and Calpain-2 are ubiquitous and have similar heterodimer structures requiring interaction with the calpain small subunit 1 (CAPNS1) and Ca2+ for activity (Ono et al. 2016). Calpain-1 requires micromolar concentrations of calcium, while Calpain-2 requires millimolar amounts of calcium (Ono et al. 2016).
