4. Materials and Methods 4.1. Experimental Materials and DesignThe experiments were conducted in the greenhouse facilities of Shandong Agricultural University and the Shandong Institute of Sericulture using the cucumber cultivar ‘Jinyan 4’. Initially, cucumber seeds were sterilized in a 0.3% hypochlorite solution for 10 min before being thoroughly rinsed with distilled water. The seeds were then germinated at a stable temperature of 28 °C.

Plasmid construction and cell transfectionAll plasmids were constructed with restriction-enzyme digestion and recombination methods (Vazyme). The pcDNA3.0 backbone vector was used for the construction of circGFP minigene and mCherry plasmid. For circGFP minigene, the annotated flanking sequences of circVN1R1, two split GFP fragments and an IRES element were inserted into the backbone between Hind III and Apa I.
