AbstractHIV can persist in a latent form as integrated DNA (provirus) in resting CD4+ T cells of infected individuals and as such is unaffected by antiretroviral therapy (ART). Despite being a major obstacle for eradication efforts, the genetic variation and timing of formation of this latent reservoir remains poorly understood. Previous studies on when virus is deposited in the latent reservoir have come to contradictory conclusions.

Sensitivity analysis and drug interventionHaving a detailed model of the intracellular replication of plus-strand RNA viruses, we next addressed the question of which processes shared across all viruses showed the highest sensitivity index to potential drug interventions (Fig 6). Our sensitivity analysis suggests that model parameters associated with vRNA translation () and synthesis within the RO ( and ) are highly sensitive for all viruses.

With this general formula, multiple models can be tested. If we fix b3=0, then the third phase of decay starting at time Ts is flat and remains at the value of y attained at time Ts. On the other hand, if we fix Ts at a large value (say Ts=10,000) then we never reach Ts and have the usual biphasic decay model. In this latter case, with A = 0, we can test for a single decay model, estimating only b2. Note that in the case of a single decay, b1 is not estimated.
