AbstractChromosomal instability (CIN) is common in solid tumours and fuels evolutionary adaptation and poor prognosis by increasing intratumour heterogeneity. Systematic characterization of driver events in the TRACERx non-small-cell lung cancer (NSCLC) cohort identified that genetic alterations in six genes, including FAT1, result in homologous recombination (HR) repair deficiencies and CIN.

Dear EditorThe BMJ Investigation relating to the NHS-Galleri Trial(1) raises important public health issues but is inaccurate. Of concern is the view that the trial is unethical. The trial was judged ethical by the Health Research Authority (Wales Research Ethics Committee 1: 21/WA/0141). This was not stated in the article. The claim that the trial is unethical is damaging to the conduct of the trial and was repeated on social media and in national press headlines.

Charles Swanton obtained a PhD from the Imperial Cancer Research Fund Laboratories (now the Francis Crick Institute) in 1998 and completed his medical oncology and Cancer Research UK (CRUK)-funded postdoctoral clinical scientist training in 2008. He was appointed chair in personalized cancer medicine at the UCL Cancer Institute, and consultant thoracic medical oncologist at UCL Hospitals in 2011.

Recent work proposes that high intra-tumoral heterogeneity can offset immunogenicity in sporadic MMRd, suggesting a potential mechanism of immunotherapy failure.

Air pollution is responsible for 8 million deaths a year globally, which is as many as tobacco. And air pollution isn't just causing lung cancer, it may cause other cancers too—including neck cancer and mesothelioma—and diseases such as cardiovascular disease, strokes, heart attacks, type 2 diabetes, and neurodegenerative disease. We urgently need public health measures to lower pollution levels. In the lab, we've been studying how air pollution causes lung cancer.

AbstractImmunotherapy revolutionized treatment options in cancer, yet the mechanisms underlying resistance in many patients remain poorly understood. Cellular proteasomes have been implicated in modulating antitumor immunity by regulating antigen processing, antigen presentation, inflammatory signaling and immune cell activation. However, whether and how proteasome complex heterogeneity may affect tumor progression and the response to immunotherapy has not been systematically examined.
