One cell at a timeSlide-seq, reported in 2019, involves transferring slices of tissue onto arrays of special beads, each one tagged with a DNA barcode that identifies its location in the array. The beads are 10 micrometers in diameter — about the same size as many cells in the body, so most beads only bind to the messenger RNAs from one cell. However, some beads capture the RNA of multiple cells at once. “So it meant that we don’t truly get single cell resolution with Slide-seq,” said Chen.

Statistics and reproducibilityFor 3D imaging and immunofluorescence staining experiments, each staining was repeated on at least two separate individuals or separate regions. For immunofluorescence validation studies, commercially available antibodies were used; 13 out of the 15 tissue samples were also analysed using snCv3 or scCv3. For ISH, 6 tissue samples (4 biopsies and 2 nephrectomies) were analysed.
