The γ1-3 subunits facilitate BK channel activation by shifting the voltage-dependence of channel activation toward the hyperpolarization direction by about 50-150 mV in the absence of Ca2+.

Abstract Voltage-gated potassium channels are critical in modulating cellular excitability, with Slo (slowpoke) channels forming a unique family characterized by their large conductance and dual regulation by electrical signals and intracellular messengers. Despite their structural and evolutionary similarities, Slo1 and Slo3 channels exhibit significant differences in their voltage-gating properties.

Concatenated tandem constructs have been proved to be necessary and powerful in understanding the subunit stoichiometry of K+ channel gating and regulation by constraining the stoichiometry, organization, and mutation of individual subunits.
