AbstractMost cancer cells reprogram their glucose metabolic pathway from oxidative phosphorylation to aerobic glycolysis for energy production. By reducing enzyme activity of pyruvate kinase M2 (PKM2), cancer cells attain a greater fraction of glycolytic metabolites for macromolecule synthesis needed for rapid proliferation.

AbstractBackground: CD24 plays a crucial role not only in promoting tumor progression and metastasis but also in modulating macrophage-mediated anti-tumor immunity. However, the impact of tumor CD24 on the immune landscape of the tumor microenvironment (TME) remains poorly explored. Here, we investigated the role of CD24a, murine CD24 gene, in the progression and immune dynamics of the tumor microenvironment (TME) in the 4T1 murine model of triple-negative breast cancer (TNBC).

Hs578T, MDA-MB-231-PT cells, IV2 cells [23], and LC cells [19] were maintained in DMEM culture medium containing 10% fetal bovine serum, 2 mM l-glutamine, and 1% penicillin/streptomycin and cultured in a humidified incubator at 37 ℃ supplied with 5% CO2. All cell lines have tested negative for mycoplasma contamination and the 231-derived sublines have been authenticated by STR profiling (Additional file 2: Figs. S6, S7, S8).
