Editor’s summaryIn contrast with proliferating cells, the activation of the RHIM domain–containing kinase RIPK3 in neurons does not induce necroptosis but rather an antiviral transcriptional response. Kofman et al. investigated the molecular mechanisms underlying this response. In cortical neurons, the RIPK3-dependent transcriptional response required other RHIM domain–containing proteins, RIPK1 and TRIF, that are also critical for necroptosis in proliferating cells.
