CGE capillary gel electrophoresis CLuc Cypridina luciferase dsRNA double-stranded RNA IVT in vitro transcription RNAP RNA polymerase 1 INTRODUCTION The clinical success of SARS-Cov-2 vaccines established synthetic mRNA as an effective drug format, paving the way for hundreds of new mRNA-based vaccines and gene therapies to enter clinical trials.[1, 2] This has resulted in a sharp increase in global demand for mRNA production, and shifted mRNA manufacturing from a relatively niche process to...

AbstractAll mRNA products are currently manufactured in in vitro transcription (IVT) reactions that utilize single-subunit RNA polymerase (RNAP) biocatalysts. Although it is known that discrete polymerases exhibit highly variable bioproduction phenotypes, including different relative processivity rates and impurity generation profiles, only a handful of enzymes are generally available for mRNA biosynthesis.
