AbstractTauopathies are a group of age-related neurodegenerative diseases characterized by the accumulation of pathologically hyperphosphorylated tau protein in the brain, leading to prion-like aggregation and propagation. They include Alzheimer’s disease (AD), progressive supranuclear palsy (PSP), corticobasal degeneration (CBD), and Pick’s disease (PiD).

Skin samples were collected from two primary sources: (1) Cadavers with neuropathologically confirmed diagnoses of tauopathies and normal controls, and (2) Living patients with clinical diagnoses of AD and PSP. We performed RT-QuIC technique to examine the prion-like seeding activity of pathological tau. Statistical analyses were conducted to assess the sensitivity and specificity of the assay, utilizing different tau substrates to understand the impact on assay performance.
