Two independent mega-analyses across a total of 32 cohorts and >3,400 patients and >2,800 controls worldwide revealed that persons with major depressive disorder (MDD) show older appearing brains compared to non-depressed peers. However, the biological mechanisms underlying the age-related structural brain alterations remain unknown. Here, we examined whether variations in the “brain age gap” were linked to variations in polygenic risk for several depression-related traits.
