The electrochemical modulation of single molecule fluorescence Herein is presented our discovery of an electrochemical approach for modulating fluorophores between fluorescent bright states and dim states. We demonstrate how to effectively modulate the fluorescent intensity of organic dye-labelled cell samples on an indium tin oxide surface using electrochemistry with redox-active mediators present in an oxygen scavenger buffer.

1 INTRODUCTION Jellies are gelatinous zooplankton belonging to the phyla Cnidaria and Ctenophora. The prevalence of jellyfish blooms has received extensive concern, and blooming jellyfish were recorded as causing blockage of nuclear power plants (Wang et al., 2023), threatening the survival of other marine animals (Baxter et al., 2011) and imposing huge economic losses on fisheries and tourism (Baumann & Schernewski, 2012; Conley & Sutherland, 2015).

AbstractStochastic optical reconstruction microscopy (STORM) allows widefield imaging with single molecule resolution through calculating the coordinates of individual fluorophores from the separation of the fluorophore emission in both time and space. Such separation is achieved by photoswitching the fluorophores between a long lived OFF state and an emissive ON state.
