The African swine fever virus RNA polymerase serves as a crucial multifunctional protein complex responsible for genome transcription and regulation. Therefore, it is essential to investigate its structural and functional characteristics for the prevention and control of African swine fever.

AbstractTo solve three-dimensional structures of biological macromolecules in situ, large numbers of particles often need to be picked from cryo-electron tomograms. However, adoption of automated particle-picking methods remains limited because of their technical limitations. To overcome the limitations, we develop DeepETPicker, a deep learning model for fast and accurate picking of particles from cryo-electron tomograms.

AbstractThe propulsion for mammalian sperm swimming is generated by flagella beating. Microtubule doublets (DMTs) along with microtubule inner proteins (MIPs) are essential structural blocks of flagella. However, the intricate molecular architecture of intact sperm DMT remains elusive. Here, by in situ cryo-electron tomography, we solved the in-cell structure of mouse sperm DMT at 4.5–7.5 Å resolutions, and built its model with 36 kinds of MIPs in 48 nm periodicity.

The chiral and deconfinement phase transitions under rotation have been simultaneously investigated in the Polyakov-Nambu-Jona-Lasinio (PNJL) model. An interesting observation has been found that the chiral phase transition is catalyzed and the deconfinement phase transition is decelerated by rotation, therefore a chiral symmetric but confined phase is induced by rotation, which indicates that chiral dynamics and gluon dynamics can be split by rotation.

Background. This study employed integrated network pharmacology approach to explore the mechanisms underlying the protective effect of colchicine against acute lung injury (ALI). Methods. We analyzed the expression profiles from 13 patients with sepsis-related ALI and 21 controls to identify differentially expressed genes and key modules. ALI-related genes were curated using databases such as DisGeNET, Therapeutic Target, and Comparative Toxicogenomics Database to curate ALI-related genes.
