From December 2019 to December 2022, a total of 218 patients with CSVD and 103 healthy controls (HCs) were included at Shandong Provincial Hospital in this study. All subjects signed informed consent prior to the study, which was approved by the Shandong Provincial Hospital Affiliated to Shandong First Medical University Subcommittee on Human Studies Institutional Review Board.

1 INTRODUCTION Cerebral small vessel disease (CSVD) is a neurodegenerative disease characterized by significant age-related changes, predominantly affecting neurons in specific brain regions. Diagnosis primarily relies on clinical symptoms and neuroimaging. However, early disease diagnosis is challenging due to variable clinical presentations that often overlap with symptoms of other degenerative diseases.

1 INTRODUCTION Osteoporosis (OP), a metabolic bone disorder, is highly prevalent among middle-aged and older adults and manifests as bone loss or loss of bone microarchitecture, augmenting bone fragility and fracture risk. Emerging evidence from a meta-analysis suggests that the global incidence of OP and bone loss, as described by the World Health Organization (WHO), was 19.7% (95% CI: 18.0%–21.4%) and 40.4% (95% CI: 36.9%–43.8%), respectively.

To explore intermediate components that modulate neuroinflammation-related depression, we exposed adult male mice to LPS intraperitoneally for 10 days to mimic depression. (Fig. S1a). Validation of mice with depressive-like phenotypes was achieved by the use of tail suspension test (TST) and forced swimming test (FST) (Fig. S1b, c). We found that Pdcd4 mRNA and protein levels are increased in the prefrontal cortex (PFC) but not in the hippocampus (HIP) of the LPS-induced depressive mice (Fig. 1a–c).
