AbstractThe genetic architecture of white matter lesions (WMLs) in Asian populations has not been well-characterized. Here, we performed a genome-wide association study (GWAS) to identify loci associated with the WML volume. Brain MRI and DNA samples were collected from 9479 participants in the Japan Prospective Studies Collaboration for Aging and Dementia (JPSC-AD). The GWAS confirmed three known WML-associated loci (SH3PXD2A, GFAP, and TRIM47).

Private enterprises offer preimplantation genetic testing with polygenic scores to select embryos with ‘desirable’ potential. In silico simulations using biobank resources show that the selected embryo would rely substantially on the choice of polygenic score method and randomness in score construction, which raises ethical concerns.

ParticipantsWe used the follow-up data of the BBJ project12. This follow-up survey from 2010 to 2014 included 141,612 participants with 32 systemic diseases at the time of participation13. We included participants genotyped by DNA genotyping array. We excluded individuals <20 years old and participants with amyotrophic lateral sclerosis. After standard quality control of genotype data (described below), we included 137,693 individuals in the analysis.
