Confocal MicroscopyMODE-K cells were plated onto glass slides, treated with 4 mM FITC-labeled stachyose for 0-4 h, and fixed in 4% paraformaldehyde for 0.5 h. Following fixation, cells were permeabilized in acetone at 4 °C for 5 min and stained. Formalin-fixed small intestinal tissues were embedded in paraffin and 10 μm sections were cut and stained. Nucleus were stained with DAPI and cytoskeleton stained with Rhodamine-phalloidin. Images were acquired by confocal microscopy with a 63X objective.

In this study, we compiled genome size estimates for 35 Ensifera species, including seven measured via FCM (Supplementary Table S2) and 28 from previous studies [24]. For five of the seven species, genome sizes were measured by sex; however, for Vescelia pieli and Gampsocleis sedakovii, only one sex was sampled due to limited availability.

Two-year-old healthy CGSs were obtained from Hanzhong Ancient Giant Salamander Co., Ltd (Hanzhong, China).
