AbstractSex differences, driven in part by steroid hormones, shape the structure and function of the brain throughout the lifespan and manifest across brain health and disease. The influence of steroid hormones on neuroplasticity, particularly in the adult hippocampus, differs between the sexes, which has important implications for disorders and diseases that compromise hippocampus integrity, such as depression and Alzheimer disease.

Ovarian hormones such as estrogens and progesterone fluctuate across the female lifespan with natural declines occurring at menopause, typically between the ages 45 and 55. The cessation of ovarian function during the menopausal transition has been linked to an array of neural changes 1, including a decline in brain glucose metabolism 2, reductions in gray matter (GM) and white matter (WM) volume 3–6, and increased amyloid-beta deposition 7 as well as WM lesions 8.

We launched the ENIGMA-Neuroendocrinology working group with the aim to address knowledge gaps about the role of sex hormones in the brain, which lead to prevalent sex- and gender-based health disparities in biomedical research. We approach this by adopting a lifespan perspective to explore the interplay of hormonal dynamics and mental health in the brain.

AbstractThe placenta plays a role in fetal brain development, and pregnancy and birth complications can be signs of placental dysfunction. Birth asphyxia is associated with smaller head size and higher risk of developing schizophrenia (SZ), but whether birth asphyxia and placental genomic risk factors associated with SZ are related and how they might impact brain development is unclear.
