α-Synuclein (αSyn) pathology is a major feature of Parkinson's disease (PD), and the occurrence of pathogenic variants in the SNCA gene1-12 or multiplication of its locus13, 14 supports a possible causative role of αSyn in PD. Yet the nature of this role has long been a matter of debate due to unclear αSyn-mediated mechanisms of cell dysfunction, degeneration, and death.
