Introduction Kinetochores assemble at centromeres by interacting with a variant histone H3 known as centromere protein A (CENP-A) in vertebrates and as Cse4 in Saccharomyces cerevisiae and other point-centromere yeast. CENP-A and Cse4 deviate from canonical histone H3 at residues interspersed in the histone core and in the sequence and length of the N-terminal segment (Fig 1A).
