Editor’s summaryNeuroblastoma is a pediatric cancer that originates from progenitor cells arising from the neural crest. A subtype of this cancer is characterized by the presence of immature mesenchymal (MES) cells, which are resistant to standard treatments, including retinoic acid therapy. Nguyen et al. screened small-molecule activators of the microRNA miR-124, which drives differentiation in neuroblastoma cells, and identified PP121, a drug that inhibits tyrosine and phosphoinositide kinases.

AbstractHeart failure involves metabolic alterations including increased glycolysis despite unchanged or decreased glucose oxidation. The mitochondrial pyruvate carrier (MPC) regulates pyruvate entry into the mitochondrial matrix, and cardiac deletion of the MPC in mice causes heart failure. How MPC deletion results in heart failure is unknown.
