The present analysis focused on two population-based Cooperative Health Research in the Region of Augsburg (KORA) cohorts for discovery and validation. The inclusion and exclusion criteria are illustrated in Additional file 1: Figure S1. The KORA S4 study enrolled 4261 participants from 1999 to 2001 [21]. The KORA S4 discovery sample used in the present analysis was restricted to those aged 55–74 with available proteomics data (n = 1653).

All animal experiments were performed in accordance with the ARVO statement on the use of animals in ophthalmic and vision research and were approved by the local authorities (Regierung von Oberbayern, ROB-55.2-2532.Vet_02-18-143). Mice were kept under standard conditions on a 12 h light/ dark cycle with access to water and food ad libitum. Pde6bSTOP mice were generated in the Barbara & Donald Jonas Stem Cells Laboratory, Columbia University, USA [77].

Study populationThis study was based on the Cooperative Health Research in the Region of Augsburg (KORA) S4/F4/FF4 cohort study [17, 18]. A total of 4261 adults, aged 25–74 years, were included at baseline between 1999 and 2001 (S4) in Germany. Follow-up examinations were conducted after 7 years (F4) and after 14 years (FF4). The present analysis was restricted to participants aged 55–74 years at S4 (N = 1653). The flow chart of study participants is presented in Fig. 1.

To date >170 post-transcriptional modifications have been reported. They can affect RNA stability, translation and localization, and hence constitute an important layer of gene regulation (1–3). Amongst the different types of transcripts, tRNA is the most heavily modified RNA species, with an average of 13 modifications per tRNA molecule (4–6). These modifications play a role in tRNA stability and folding, although their exact biological function often remains elusive.
