Chimeric antigen receptor (CAR)-T cells have revolutionized the treatment of haematological malignancies. However, challenges — such as relapse owing to insufficient persistence of CAR-T cells — remain. Now, writing in Science Translational Medicine, Bailey et al. have demonstrated that knocking out the interferon-gamma (IFN-γ) receptor (IFN-γR) in CAR-T cells enhances their efficacy and persistence in models of haematological and solid tumours.

AbstractMaintaining pH homeostasis is critical for cellular function across all living organisms. Proton-sensing G protein-coupled receptors (GPCRs), particularly GPR4, play a pivotal role in cellular responses to pH changes. Yet, the molecular mechanisms underlying their proton sensing and activation remain incompletely understood. Here we present high-resolution cryo-electron microscopy structures of GPR4 in complex with G proteins under physiological and acidic pH conditions.

The landscape of cannabis use has changed rapidly, with over 40% of adults aged 19 to 30 years using cannabis in the past year. Of great concern is the rising prevalence in pregnant women, fueled by the legalization and its public perception of safety. However, emerging evidence from animal/in vitro and human studies suggests that prenatal cannabis exposure influence maternal physiology and fetal development, with impact on short- and long-term outcomes. While cannabis exposure is well-known for its impacts on cognitive functioning and neurodevelopment, it also affects other organ systems, leading to long-term cardiovascular, hematologic, gastrointestinal, growth, and endocrine/metabolic dysregulations. This narrative mini-review describes the historical background of cannabis, and provides an overview of how prenatal cannabis exposure affect maternal physiology and fetal development across multiple organ systems. We aim to provide clinicians with a practical understanding of the biological and developmental impact of prenatal cannabis exposure to support evidence-informed counseling during pregnancy and lactation.

AbstractSevere cutaneous adverse drug reactions (SCARs) are life-threatening diseases, which are associated with human leukocyte antigen (HLA) risk variants. However, the low positive predictive values of HLA variants suggest additional factors influence disease susceptibility. Using dapsone hypersensitivity syndrome (DHS) as a paradigm for SCARs, we show that the DHS patients harbor a sex-related global reduction in blood NK cells, contributing to the higher incidence of reactions in females.

AbstractMany relationships between pathogens and human disease are well-established. However, only a small fraction involve diseases considered non-communicable (NCDs). In this study, we sought to leverage the vast amount of newly available electronic health record data to identify potentially novel pathogen-NCD associations and find additional evidence supporting known associations.
