AbstractPolygenic scores (PGS) enable the exploration of pleiotropic effects and genomic dissection of complex traits. Here, in 421,889 individuals with European ancestry from the Million Veteran Program and UK Biobank, we examine how PGS of 17 neuropsychiatric traits are related to membership in 22 broad professional categories.

AbstractCannabis use disorder (CUD) and cannabis use (CU) are prevalent conditions co-occurring with attention-deficit hyperactivity disorder (ADHD). Here we report results from a cross-disorder genome-wide association study of ADHD and CUD or CU. We identified 36 concordant genome-wide significant loci for ADHD–CUD and ten loci for ADHD–CU. DRD2 was identified as an ADHD–CUD risk gene.

AbstractMajor depressive disorder (MDD) and cardiovascular disease (CVD) are often comorbid, resulting in excess morbidity and mortality. Here we show that CVDs share most of their genetic risk factors with MDD. Multivariate genome-wide association analysis of shared genetic liability between MDD and atherosclerotic CVD revealed seven loci and distinct patterns of tissue and brain cell-type enrichments, suggesting the involvement of the thalamus.

Here we conducted a large cross-ancestry meta-analysis of PAU in 1,079,947 individuals (European, N = 903,147; African, N = 122,571; Latin American, N = 38,962; East Asian, N = 13,551; and South Asian, N = 1,716 ancestries).
